From the fruits of Cordia dichotoma, 11 new phenolic compounds, dichotomins A-K, were isolated, together with 19 known compounds. Through the analysis of detailed NMR data and HRESIMS data, the planar structures of all compounds were confirmed. Using NMR calculations, the absolute configuration of dichotomins A-K was elucidated by comparing their observed and computed electronic circular dichroism (ECD) spectra. Dichotomin H (8) and dichotomin I (9) were determined as two pairs of enantiomers. The enantiomers of compounds 8 and 9 were separated using chiral-phase high-performance liquid chromatography (HPLC), and the stereostructure of each enantiomer was determined by similarly calculating the ECD. Compounds 3, 5, 7, 17, 18, 23−25, and 27−30 increased glucose uptake by 1.04-to 2.85-folds at concentrations of 30 μg/mL. Further studies revealed that compounds 3 and 5 had a moderate effect on glucose transporter 4 (GLUT4) translocation activity in L6 cells. At 30 μg/mL, compound 3 significantly enhanced AMPK phosphorylation and GLUT4 expression. As a whole, compound 3 has the potential to be a drug candidate for the treatment of type 2 diabetes mellitus (T2DM).

Cordia subcordata trees or shrubs, belonging to the Boraginaceae family, have strong resistance and have adapted to their habitat on a tropical coral island in China, but the lack of genome information regarding its genetic background is unclear. In this study, the genome was assembled using both short/long whole genome sequencing reads and Hi–C reads. The assembled genome was 475.3 Mb, with 468.7 Mb (99.22%) of the sequences assembled into 16 chromosomes. Repeat sequences accounted for 54.41% of the assembled genome. A total of 26,615 genes were predicted, and 25,730 genes were functionally annotated using different annotation databases. Based on its genome and the other 17 species, phylogenetic analysis using 336 single-copy genes obtained from ortholog analysis showed that C. subcordata was a sister to Coffea eugenioides, and the divergence time was estimated to be 77 MYA between the two species. Gene family evolution analysis indicated that the significantly expanded gene families were functionally related to chemical defenses against diseases. These results can provide a reference to a deeper understanding of the genetic background of C. subcordata and can be helpful in exploring its adaptation mechanism on tropical coral islands in the future.

The study aimed to characterize hydro-methanolic (25%) extracts of Justicia adhatoda L. (stem and flower) and Cordia dichotoma Frost. (Stem and fruit) and evaluate the in silico thrombolytic properties of the major phytocompounds present in the plants. In the preliminary fluorescence imaging through treatment by different reagents, both plants were found to be pharmacologically active. Further qualitative screening of phytochemicals, spectroscopy-based techniques, namely, UV-Vis Spectroscopy and FTIR, revealed various classes of compounds such as polyacetylenes, aglycones, quercetin, anthocyanins, anthraquinones, alkaloids, chalcones and aurones, flavanols, carotenoids, and flavanones. Further, by the application of Thin Layer Chromatography, phenols and flavonoids, namely Catechol, Kaempferol, Quercetin, and Lutein, along with other compounds like Chlorophyll b, Glutamic Acid, and Tryptophan were identified from the extracts. Finally, in the molecular docking study, three compounds, Datiscoside and Robinin of C. dichotoma and Daucosterol of J. adhatoda showed high binding energies (-10.224, -9.547 and -9.262 kcal mol-1 respectively) towards the G-protein coupled thrombotic platelet aggregation receptor P2Y1 when compared to that of the control MRS2500 (-7.148 kcal mol-1).
