Phenotyping of macrophages with monoclonal antibodies in endomyocardial biopsies as a new approach to

Abstract: Cryostat sections of endomyocardial biopsies from 53 patients (mean age 41 +/- 5 years, 38 male and 15 female) clinically indicated to suffer from myocarditis were stained using monoclonal antibodies against subpopulations of T-lymphocytes and macrophages and with polyclonal rabbit-anti-human sera marking two calcium-binding proteins expressed by monocytes and macrophages appearing in inflammatory sites only. No inflammatory infiltrate cells were found in 13 cases (25%). Mononuclear cell infiltrates were prese… Show more

“…Consequently these patients were classified as patients with idiopathic dilated cardiomyopathy. These criteria of diagnosing myocardial inflammation were based on published results (20,23,26,27,49) and on our own control group (n = 85) (19) in which mean CD3 T-lymphocyte count/HPF in normal myocardial tissue were 0.7 (range 0.0-1.4). In addition, a subgroup analysis was performed of patients with a CD3 T-lymphocyte count ≥ 3 CD3 T-lymphocytes/HPF (≥ 11…”

“…S100A8/S100A9 complex is a very early serum marker suitable for monitoring of acute rejection with high sensitivity (67%) and specificity (100%) (Burkhard et al 2001). S100A8-and S100A9-expressing macrophages are also found during the early phase of cardiac allograft rejections (Mues et al, 1990).…”

“…MRPs are expressed primarily by circulating human neutrophils and monocytes. Moreover, MRP14 and MRP8 are expressed on activated macrophages in such acute and chronic inflammatory processes as rheumatoid arthritis [13,15], myocarditis [10], granulomatous reactions [4], cystic fibrosis [17], and Crohn's disease [19]. However, MRPs are negative on mature tissue macrophages.…”
