Phloretin Promotes Adipogenesis via Mitogen-Activated Protein Kinase Pathways in Mouse Marrow Stromal ST2 Cells

Abstract: Phloretin, a glucose transporter (GLUT) inhibitor, has pleiotropic effects. The present study examined the effects of phloretin on the commitment of marrow stromal cells to adipocytes, using the mouse marrow stromal cell line ST2. Oil red O staining showed that treatment with phloretin 10–100 µM promoted lipid accumulation. Real-time PCR showed that phloretin significantly increased the expression of adipogenic markers, including PPARγ, C/EBPα, fatty acid synthase, fatty acid-binding protein 4, and adiponectin… Show more

“…Phloretin can broadly activate the AMPK pathway in most cells, such as murine preadipocytes [33], murine lung fibroblasts [34], murine osteocytes [35], mouse marrow stromal cells [36], and human umbilical vein endothelial cells (HUVECs) [37]. In this study, the phosphorylation level of AMPK was significantly enhanced and the phosphorylation level of mTOR was significantly reduced in human activated CD4 + T cells under phloretin treatment.…”

“…Phloretin is a main compound, which belongs to the dihydrochalcone class among flavonoids and also exists in strawberries and pears. Phloretin has a wide range of biological effects, including antioxidation, anti-inflammatory, and anti-tumor properties, and it reduces vascular endothelial dysfunction and liver injury [21][22][23]. At present, phloretin and its glucoside phlorizin are generally considered health-beneficial polyphenols from apples [5].…”

“…Furthermore, the inhibition of JNK using phloretin, a glucose transporter (GLUT) inhibitor, induced adipogenesis in marrow stromal cells [36]. In addition, treatment with the JNK inhibitor SP600125 significantly increased lipid accumulation in ST2 cells [36]. Other studies, however, argue that JNK is not directly involved in the differentiation process of adipocytes [72].…”

“…It has been proposed that the activation of PKA or JNK during adipocyte differentiation can lead to lipolysis and abolish adipogenesis [36,37]. However, little is known about the crosstalk between the two proteins in coordinating the hydrolysis of triglycerides and how this affects the overall differentiation process.…”
