Phospholipid Metabolism in T4 Bacteriophage-infected Escherichia coli K-12 (λ)

Abstract: Infection of Escherichia coli K-12 (X) by bacteriophage results in an altered labeling pattern of phospholipids in the host cell. Although the overall incorporation of 82Pi into phospholipids is decreased by infection, the relative amounts of phosphatidylglycerol and cardiolipin are increased. Phospholipid changes occurring at later stages in the lytic cycle of infected bacteria are more prominent than those at earlier time intervals. The uptake of 32Pi into phospholipids of cells infected with T4Bs and endoly… Show more

“…Infection with these phage is abortive if the medium contains as little as 1 mm monovalent cation; any monovalent cation inhibits, and magnesium, or to a lesser extent calcium, overcomes the inhibition (15). Cardiolipin synthesis is increased in rll-infected cells compared to controls infected with r+ (31). The increase in cardiolipin occurs too late to be considered the cause of abortive infection, however, and the similarity of ionic requirements and phospholipid synthesis between rII infection and sodium inhibition in strain A324-1 may be coincidental.…”

“…The radioactivity in each spot was determined by scintillation spectrometry as previously described (5) except that 3a70B scintillation fluid was used. The identity of the individual phospholipids was established as previously described (30).…”

“…To assure complete recovery, all of the gels in each lane were routinely assayed by this procedure. The identities of the individual phospholipids were established by the methods ofPeterson and Buller (11).…”
