Phosphonate terminated PPH dendrimers: influence of pendant alkyl chains on the in vitro anti-HIV-1 properties

Abstract: The synthesis, characterization and in vitro anti-HIV activity of a series of generation one dendrimers having phosphonate groups with pendant alkyl chains are described. The influence of the lateral alkyl chains on the biological properties was correlated to (1)H-(1)H NOESY experiments.

“…The following cationic polymers were used for transfection: linear polyethylenimine ExGen500 (Fermentas), fourth-generation polyamidoamine dendrimers PAMAM-NH 2 G4 (Dendritic NanoTechnologies) and fourth-generations phosphorus dendrimers (Phos G4) [5,12,15]. Formation of DNA-polycation complexes was conducted in 150 mM NaCl at a charge ratio of 1:1 (for PEI ExGen500 according to manufacturer's recommendation), transfection was carried out for 2 h in culture medium containing 10% serum.…”

“…Blanzat et al have evaluated anti-HIV potential of phosphorus dendrimers based on hexachlorocyclotriphosphazene or thiophosphoryltrichloride core in a series of investigations [8082]. Phosphorous dendrimers with either cinnamic acid or phosphonic acid end groups were prepared.…”

“…Thus, this series of dendrimers was deemed to be unsuitable for further development. The same group designed phosphonate terminated poly-(phosphorhydrazone) dendrimers with different pendant alkyl chains [82]. Unlike earlier investigation, no attempts were made to fabricate cataninonic dendrimers.…”
