Phosphoregulation of the septin cytoskeleton in neuronal development and disease

Abstract: Septins are highly conserved GTP‐binding proteins that oligomerize and form higher order structures. The septin cytoskeleton plays an important role in cellular organization, intracellular transport, and cytokinesis. Kinase‐mediated phosphorylation of septins regulates various aspects of their function, localization, and dynamics. Septins are enriched in the mammalian nervous system where they contribute to neurodevelopment and neuronal function. Emerging research has implicated aberrant changes in septin cyto… Show more

“…It has been a mystery why the phenotypes of septin mutants are so mild in fission yeast ever since their discoveries in the early 1990s, yet their sequences and structures are evolutionarily conserved across species (An et al, 2004;Zheng et al, 2018;Zheng et al, 2024). In human, dysregulation of septins or exocyst complex leads to severe disorders including neurological diseases and cancers (Russell and Hall, 2005;Martin-Urdiroz et al, 2016;Halim et al, 2023;Werner and Yadav, 2023). Thus, it is critical to identify the functional links between septins and exocyst complex for better understanding and treatment of related diseases.…”

“…Additionally, the increase in SEPTIN proteins in B27 treatment suggests changes in the neurodevelopment realm. SEPTIN proteins contribute to neurodevelopment and neuronal functions in the mammalian nervous system [64], with SEPTIN3 being highly expressed in the brain [65]. SEPTIN3 protein is involved in presynaptic plasticity via the cGMP/PKG pathway, and its expression is developmentally regulated as nervous system development increases [65] and it is predominantly abundant in nerve terminals and presynaptic and synaptic vesicles [65,66].…”
