Phosphorylation-Coupled Proteolysis of the Transcription Factor MYC2 Is Important for Jasmonate-Signaled Plant Immunity

Abstract: As a master regulator of jasmonic acid (JA)–signaled plant immune responses, the basic helix-loop-helix (bHLH) Leu zipper transcription factor MYC2 differentially regulates different subsets of JA–responsive genes through distinct mechanisms. However, how MYC2 itself is regulated at the protein level remains unknown. Here, we show that proteolysis of MYC2 plays a positive role in regulating the transcription of its target genes. We discovered a 12-amino-acid element in the transcription activation domain (TAD)… Show more

“…As shown in Figures 7A and 7B, after inoculation with spores of B. cinerea, the myc2 mutant exhibited disease resistance, as indicated by the smaller lesion size compared with the wild type, which is similar with previous studies demonstrating that MYC2 negatively regulates resistance against B. cinerea (Lorenzo et al, 2004;Zhai et al, 2013). The ein3 eil1 (C) Co-IP assay to verify the interaction of MYC2 with EIN3 in planta.…”

“…Surprisingly, MYC2 also acts as a negative regulator to repress JA-mediated plant resistance to necrotrophic fungi and pathogenesis-related gene expression (e.g., PDF1.2) (Anderson et al, 2004;Lorenzo et al, 2004;Zhai et al, 2013). Such MYC2-regulated susceptibility to necrotrophic fungi seems incompatible with the previously reported synergistic model (Zhu et al, 2011).…”

“…MYC2, MYC3, and MYC4 are stabilized by light, via the action of red and blue light photoreceptors, and also by JA treatment (Chico et al, 2014). Phosphorylationdependent proteolysis of MYC2 was reported to be essential for its transcriptional activity (Zhai et al, 2013). In all these cases, MYC protein degradation was mediated by the proteasome.…”

“…Considering that MYC2 regulates diverse aspects of JA responses (Boter et al, 2004;Lorenzo et al, 2004;Dombrecht et al, 2007;Zhai et al, 2013), we examined whether this master regulator is also involved in COI1-mediated regulation of flowering time. Interestingly, in both LD and SD conditions, the flowering time of the myc2-2 mutant, which harbors a T-DNA insertion that disrupts the function of MYC2 (Boter et al, 2004), was comparable to its wild-type counterpart ( Figures 1A to 1E), indicating that MYC2 is not involved in COI1-dependent regulation of flowering time.…”
