Phosphorylation-Elicited Quaternary Changes of GA Binding Protein in Transcriptional Activation

Abstract: Enrichment of nicotinic acetylcholine receptors (nAChR) on the tip of the subjunctional folds of the postsynaptic membrane is a central event in the development of the vertebrate neuromuscular junction. This is attained, in part, through a selective transcription in the subsynaptic nuclei, and it has recently been shown that the GA binding protein (GABP) plays an important role in this compartmentalized expression. The neural factor heregulin (HRG) activates nAChR transcription in cultured cells by stimulating… Show more

“…GABP is composed of two distinct proteins. One is GABP␣, which has an ets domain and not a transactivation domain, and the other is GABP␤1, which is a specific cofactor; together they form complexes for transactivation (6,12,19,41,50). Thus, to ascertain whether GABP can bind EBS-1 of the synoviolin promoter in NIH 3T3 cells, we prepared an EBS-1 probe (Ϫ83/Ϫ68) including EBS-1 (Ϫ76/Ϫ69) and then performed EMSA using this probe.…”

“…Additional mechanisms may involve posttranscriptional modification of NRF-2, which may signal up-or down-regulation of COX by affecting NRF-2's availability and ability to interact with other factors, such as NRF-1. Phosphorylation of NRF-2α, for example, increases its activation of nicotinic acetylcholine receptor promoters (Sunesen et al, 2003).…”

“…While there are sites for GABP, the supershift analysis indicates that GABP binds to a small degree in the upstream region and not at all in the downstream region. Since GABP is stimulated by motorneuron-derived factors such as neuregulins [32,33], GABP might be recruited to this region under different conditions, but our results suggest that the primary protein binding to the regions flanking the repressor E box under these conditions is NFI.…”
