Photodynamic treatment with hexyl-aminolevulinate mediates reversible thiol oxidation in core oxidative stress signaling proteins

Abstract: Photodynamic therapy (PDT) is a highly selective two-step cancer treatment involving a photosensitizer and illumination with visible light in the presence of molecular oxygen. PDT is clinically approved worldwide for treating several premalignant conditions and cancer forms, especially endoscopically accessible tumors and dermatological malignancies. PDT-mediated cytotoxicity takes place via autophagy, apoptosis and necrosis, but the exact trigger mechanisms for various death-pathways are still unknown. PDT in… Show more

“…Thus, the ebselen effect on 14-3-3 turnover and consequent alteration of the PPI network is also expected to be relevant for cancer treatment. Actually, cysteine residues in 14-3-3 are targets of photodynamic light cancer therapy mediated by reactive oxygen species, where oxidization of these residues is expected to mediate the apoptosis triggered by such treatment (Helander et al 2016). Our work,…”

“…HAL is a lipophilic photosensitizer with antibacterial properties generally used in bladder cancer diagnostics ( 7 9 ). HAL-mediated antifungal photodynamic therapy (aPDT) leads to reactive oxygen species production and antimicrobial activity without inducing drug resistance ( 10 ).…”

“…HAL offers the advantages of low therapeutic concentrations, few side effects, and better economic value (Neittaanmäki-Perttu et al, 2016). Besides, HAL-mediated PDT has contributed to the diagnosis and treatment of tumors and skin diseases, such as bladder cancer (Helander et al, 2016) and solar keratosis (Neittaanmäki-Perttu et al, 2017). However, its efficacy in biofilm elimination, which is crucial to the treatment of cutaneous candidiasis, has not yet been studied.…”
