Photogeneration and Quenching of Reactive Oxygen Species by Urocanic Acid

Abstract: Urocanic acid, UCA, is characterized by two electronic transitions in the UV-B (280-320 nm) which comprise its broad absorption spectrum and give rise to wavelength-dependent isomerization quantum yields. The absorption spectrum of UCA extends into the UV-A (320-400 nm). Given the UV-A component of sunlight is significantly greater than the UV-B component it is hypothesized even weak UV-A photochemistry of UCA could be important for in vivo responses to UV radiation. Degenerate pump-probe experiments performed… Show more

“…The link between FLG and UV-B bioavailability can be explained by the proteolytic processing of FLG into one of its constituent amino acids, histidine, followed by the deimination of histidine by the enzyme, histidine ammonia-lyase (histidase) into the carboxylic acid, trans-urocanic acid (t-UCA) (Scott, 1981; Brown and McLean, 2012). t-UCA is a key endogenous sunscreen of the stratum corneum (Kripke, 1984), with an action spectrum in the UV-B spectrum (Brookman et al, 2002; Haralampus-Grynaviski et al, 2002; McLoone et al, 2005). Moreover, FLG knock-down results in sub-normal levels of t-UCA and an increased susceptibility to UV-B-induced apoptosis (Mildner et al, 2010).…”

“…Exogenous agents (70) and endogenously occurring compounds including porphyrins, flavins, DNA bases, or amino acids and their derivatives like urocanic acid are considered to act as photosensitizing molecules (78). According to the postexcitational chemistry of photooxidation, the processes are assigned to either Type I or Type II photooxidation reactions (38,56,64,124).…”

“…One protein has just emerged as a candidate to explain enhanced VD3 production--the stratum corneum structural protein filaggrin (FLG) (Thyssen et al, 2014). The resulting t-UCA is a key endogenous sunscreen of the stratum corneum (Kripke, 1984) with an action spectrum in the UV-B spectrum (Brookman et al, 2002;Haralampus-Grynaviski et al, 2002;McLoone, Simics, Barton et al, 2005).
