Photosensitization of CdSe/ZnS QDs and reliability of assays for reactive oxygen species production

Abstract: CdSe/ZnS quantum dots (QDs) conjugated to biomolecules that can act as electron donors are said to be "photosensitized": that is, they are able to oxidize or reduce molecules whose redox potential lies inside their band edges, in particular molecular oxygen and water. This leads to the formation of reactive oxygen species (ROS) and phototoxicity. In this work, we quantify the generation of different forms of ROS from as-synthesized QDs in toluene; water-solubilized, unconjugated QDs; QDs conjugated to the neur… Show more

“…The protective effect of the antioxidant ERT and the photo-inducible enhancement of QDsN genotoxicity confirmed this hypothesis. The photo-genotoxic properties of QDs have already been shown by Anas et al [14] and their effects on the metabolism of mammalian cells have been demonstrated to be proportional to the amount of ROS generated [49]. In vitro studies have also shown that nanoparticles could deplete endogenous antioxidants, alter mitochondrial function and produce oxidative damage to lipids and DNA [50,51].…”

“…To increase the efficiency of wide bandgap semiconductors for the visible light harvesting, different narrow bandgap semiconductors have been introduced as photosensitizers like InAs [132], CdS [133][134][135], CdSe [136][137][138][139][140], InP [141], PbS, and so forth. The nanoheterostructures which are also called combined nanocrystals show improved property which is distinct from that of any component in them.…”

“…Cooper et al studied the potential of CdSe/ZnS QDs for ROS production (Cooper et al 2010) ROS assays in solution were performed by using sensor green while ROS generation inside cells were done by using 5-(and 6-)-chloromethyl-2¢,7¢-dichlorodihydrofluorescein diacetate, acetyl ester (Fig. 37).…”
