Phylogenetic relationship of China Dendrobium species based on the sequence of the internal transcribed spacer of ribosomal DNA

Abstract: The genetic relationship of 36 Dendrobium species in China was determined based on sequence analysis of the internal transcribed spacer (ITS) region of ribosomal DNA. Aligned sequences of the complete ITS region obtained from the 36 Dendrobium species and 2 outgroup species (Epigeneium amplum and Epigeneium nakaharaei) by using PCR amplification and direct DNA sequencing. The nrDNA ITS1 of Dendrobium was 225 -234 bp and ITS2 was 239 -248 bp. Phylogenetic tree was constructed, and seven main clusters were gener… Show more

“…D. fimbriatum and D. chrysotoxum have a close phylogenetic relationship [49], while in the present study, the two species were colonized by different xylariaceous fungi, indicating that different host plants in the same habitat might have preference and selectivity for their fungal partner.…”

“…In fact, ITS sequences were previously used to generate the first phylogeny of Rubus, which generally corresponded to their biogeography and polyploid, but not to their conventionally important morphological characters [19]. Our study has been congruent with the previous reports of Yuan et al [20] and Trung et al [14], who documented the relationship of Dendrobium species based on sequences characterization of the rDNA ITS region.…”

“…However, differences between classical and molecular approaches were also reported in studies of Dendrobium spp. found in Taiwan (Tsai et al, 2004) and China (Yuan et al, 2009). The orchids of sections Breviflores and Pedilonum were placed in the same cluster as section Dendrobium (cluster I in Fig.…”

“…In Dendrobium, the molecular phylogeny of several species has been studied using nucleotide sequences of both rDNA ITS (Burke et al, 2008;Tsai et al, 2004;Yuan et al, 2009) and the matK gene (Yukawa et al, 2000). However, the number of species included in each study was very limited owing to the range of species distributed in different regions (Australia, China and Taiwan).…”
