Phylogeography of the Northern Atlantic species Chondrus crispus (Gigartinales, Rhodophyta) inferred from nuclear rDNA internal transcribed spacer sequences

Abstract: Eighteen isolates of the red algae Chondrus crispus were collected from Northern Atlantic sites, together with C. ocellatus, C. yendoi and C. pinnulatus from the North Pacific. The nuclear rDNA internal transcribed spacer (ITS) was sequenced and compared, spanning both the ITS regions and the 5.8S rRNA gene. Percentage of nucleotide variation for C. crispus ranged from 0.3% to 4.0%. Phylogenetic analyses were performed using maximum parsimony (MP), neighbor-joining (NJ) and minimum evolution methods. They show… Show more

“…Two primer sets, Cox3F (5 0 -AHRGTTCAACGA-CAYCCATTTC-3 0 ; designed in this study) and CAR4A (5 0 -CCCCACCARTAWATNGTNAG-3 0 ; Kogame et al, 2005), ITS2FC2 (5 0 -TTGTCGGGGAGGAGGAGG-3 0 ; Cheang et al, 2010) and 25BR2 (5 0 -TCCTCCGCTTAGTATATGCTTAA-3 0 ; Yoshida et al, 2000), were used to amplify mitochondrial cox3 and nuclear ITS2 respectively. PCR, electrophoresis and sequencing were conducted following our previous protocols (Hu et al, 2007(Hu et al, , 2011.…”

“…Finally, restricted gene flow among C. crispus populations makes it potentially easier to follow the genetic signature of post-Pleistocene recolonization (Wang et al 2008). One previous study interpreted the contemporary distribution of C. crispus in the North Atlantic based on the theory of continental drift (Hu et al 2007b). The small sample size for each locality reduced the validity of this hypothesis.…”

“…For example, interfertility was demonstrated in Chondrus crispus from the east and west Atlantic coasts (Guiry, 1981(Guiry, , 1992 based on the ITS1 sequence data, the genetic distance was not to be correlated with the geographic distance at all, and hence, multiple transatlantic dispersal was suggested (Hu et al, 2007). Interfertility between geographically distant populations was also demonstrated in Bostrychia radicans from both sides of South America (Pacific Mexico vs. Venezuela, or Peru vs. Brazil), and there were only a few site changes at the Rubisco spacer plus the flanking genetic region among these interfertile strains (Zuccarello and West, 1997).…”
