Physical and genetic mapping of the genomes of five Mycoplasma hominis strains by pulsed-field gel electrophoresis

Abstract: We present the complete maps of five Mycoplasma hominis genomes, including a detailed restriction map and the locations of a number of genetic loci. The restriction fragments were resolved by field inversion gel electrophoresis or by the contour-clamped homogeneous-electric-field system of pulsed-field gel electrophoresis. All

“…horninis PG21. However, DNA probes from the gene (termed p120) encoding P120 hybridized to DNA from all 19 M. horninis strains including PG21, and on the chromosomal maps of five M. horninis strains (PG21,4195,132,93 and 7488) the pl20 gene was located at similar positions close to the gyrB gene (Ladefoged & Christiansen, 1992). To study whether this variable, surface-localized protein was recognized by the human humoral immune response we studied this system in detail.…”

“…This can be explained by the fact that these sites were mainly located in the vicinity of the rrn operons which are known to have a higher G+C content -47 mol% in the case of the FD-P 16S rDNA, as is the case for other mollicute genomes (Barré et al, 2004;Razin et al, 1998;Sirand-Pugnet et al, 2007). The concentration of G+C-rich restriction sites in the vicinity of the rrn operons has also been observed on physical maps of Mycoplasma hominis (Ladefoged & Christiansen, 1992) and Spiroplasma citri strains (Ye et al, 1992). 51/37 600 bp, i.e.…”

“…In addition, the Mycoplasma mycoides subsp. mycoides Y and M. hominis physical maps showed frequencies of restriction sites containing one A and one T (BamHI, KpnI, SalI and XhoI) six to ten times lower than the theoretical ones (Ladefoged & Christiansen, 1992;Pyle & Finch, 1988). This phenomenon could be due to methylation of the majority of these restriction sites.…”
