“…ASL, with a pKa of 6.7, actively loaded with ammonium sulphate into liposomes, presumably falls into this category. This is supported by the fact that during drug loading (See et al, 2014), ASL with <10% unionized form could be rapidly loaded into the liposomes.…”

“…Then the bags were immersed in 500 ml of release medium in different beakers on a shaking water bath (GLS aqua 18 plus, Grant Instruments, UK) preset at 37 o C. At each pre-determined time interval, samples (0.1 ml of ASL-L suspension) were withdrawn from the dialysis bags. ASL in the liposomes was extracted by dissolving the liposomes with acetonitrile and the drug concentration determined by a validated stability-indicating HPLC method (See et al, 2014). The release method capable of identifying the 'leaky' liposomes was considered to be bio-relevant, and used in the sequential release studies.…”

“…ASL was administered intravenously as an isethionate salt, which is lipophilic (log P value ≈ 3.0 at pH 7) and moderately soluble in water (the solubility is ∼1 mg/mL, pH is ∼4.5; See et al., 2014 ). Our previous studies showed that post-injection drug precipitation upon dilution of body fluid and drug irritancy were probably the main reasons for phlebitis with ASL (See et al., 2014 ; Zhang et al., 2015a ). Different strategies using liposome carriers have been attempted to improve phlebitis caused by ASL (Zhang et al., 2015a , b , 2016 ).…”

“…Marchetti 2014), such as Poloxamer 188 (P188) modified ASL-loaded long-circulating liposomes, which had a prolonged half-life but no drug precipitation compared with ASL solution (See, Zhang, Liu, Svirskis, Baguley, Shaw, Wang andWu 2014, Zhang, Wang, See, Shaw, Baguley, Liu, Amirapu andWu 2015). This way, ASL-liposome may act as an alternative formulation to prevent phlebitis during i.v.…”
