Phytase fromAspergillus niger

Abstract: 132 microorganisms, isolates from soil and decayed fruits, were tested for phytase production. All isolates intensively producing active extracellular phytase were of fungal origin. The most active fungal isolates with phytase activity were identified as Aspergillus niger. At the end of the growth phase, the extracellular phytase activity produced by A. niger strain 92 was 132 nkat/mL, with strain 89 it was 53 nkat/mL. In both strains the extracellular enzyme activity exhibited two marked activity optima at pH… Show more

“…Moreover in poultry the gut pH varies from 2.5 to 6.0, thus phytase active and stable in acidic environment is highly preferred (Radcliffe et al 1998). Among fungi many Aspergilli (Ullah 1988;Volfova et al 1994;Banerjee 2002, 2004;Vohra and Satyanarayana 2003;) are known to be active phytase producers. As Aspergillus niger is Generally Recognised as Safe (GRAS) it is frequently used in food and feed applications.…”

“…The phytase activity in the ruminal strains was found to be cell-associated, even though substantial phytase activity was found in cell-free ruminal fluid, and phosphate was released from phytate into the culture fluid by pure cultures (Yanke et al 1998). Such cell-associated activity is in direct contrast to the extracellular phytase activity reported with Bacillus subtilis (Shimizu 1992) and Aspergillus niger (Volfová et al 1994). However, native phytase activity is expressed periplasmically in E. coli in the stationary phase, particularly under anaerobic conditions (Greiner et al 1993).…”
