Pigment Epithelium-Derived Factor as a Possible Treatment Agent for Choroidal Neovascularization

Abstract: Choroidal neovascularization (CNV) is a sight-threatening disease and is characterized by the formation of pathological neovascularization in the choroid which extends into the subretinal space. Exudative age-related macular degeneration (AMD) is the formation of CNV in the macular area which leads to irreversible blindness. Continuous leakage and hemorrhage of the CNV lesion may eventually result in scarring or later fibrosis, which could result in photoreceptor cell atrophy. The current strategy for treating… Show more

“…As a first-line therapy, intravitreal injection of antivascular endothelial growth factor (VEGF) agents is effective in patients with nAMD because it targets pathological CNV (Kovach et al, 2012;Ferrara and Adamis, 2016). However, the limitations of anti-VEGF therapy should not be ignored, such as the side effects of the injection (Xi, 2020) and the unsatisfactory duration of the therapeutic effect (Ehlken et al, 2019). In addition, long-term use of anti-VEGF therapy may lead to serious economic burdens, especially in developing countries and regions (Ruiz-Moreno et al, 2021).…”

“…RPE cells produce a range of angiogenic factors, including VEGF and PEDF, which are strongly related to the development of chorioretinal diseases [ 7 ]. VEGF is a key factor promoting angiogenesis and vascular permeability [ 13 ]. It is a mitogen and chemokine of vascular endothelial cells responsible for cell division and proliferation.…”

“…So far, the key role of these cytokines has been confirmed in the pathogenesis of age-related macular degeneration (AMD) [ 8 , 9 , 10 , 11 ]. Importantly, AMD and CSC share some similarities, such as disturbances in the levels of proangiogenic and antiangiogenic factors [ 12 , 13 ] and the presence of hyperpermeability. Moreover, they both can be complicated by choroidal neovascularization (CNV).…”
