Pigmented median raphe cyst of the penis with consideration of the possible mechanism of melanocytic colonization: A case report

Abstract: Median raphe cyst is a rare lesion located on the median raphe. The cyst wall is lined by cuboidal to columnar cells, transitional (urothelial) cells, stratified squamous cells or a mixture of these. The normal urethral mucosa and the median raphe cyst usually lack melanocytes and/or melanin pigment. However, albeit extremely rare, the presence of melanin pigment and/or melanocytes in median raphe cyst, namely pigmented median raphe cyst, has been previously reported. The current case report presents the sixth… Show more

“…There is also evidence that most of these factors (LTC4, bFGF, ET‐1, SDF‐1) are expressed by meningiomas, having a role in tumorigenesis and/or angiogenesis . However, tumors in different locations may have alternate mechanisms of melanocyte colonization . We immunostained for several molecules that have been reported to affect melanocyte migration .…”

“…[20][21][22][23] However, tumors in different locations may have alternate mechanisms of melanocyte colonization. 24 We immunostained for several molecules that have been reported to affect melanocyte migration. [17][18][19][20][21][22][23] We show that the receptor CD117 (c-Kit) is expressed in a subset of the recurrent tumor's melanocytes and that its ligand, SCF (KITL), is diffusely expressed in the meningioma cells.…”

“…Although extremely rare, MRCs containing melanin pigment and/or melanocytes (i.e. pigmented MRCs) have been reported, with six cases to date having been published 4,5,10,14,15 . Given that the cyst of our patient was lined mostly by non‐keratinizing stratified squamous epithelium and partially by stratified columnar epithelium (Fig.…”

“…MRCs are rare benign lesions of the male genitalia that can develop at any site along the midline from meatus to anus (i.e. parameatus, glans penis, penile shaft, scrotum, perineum, or perianal) 1–19 . The condition was first described by Mermet in 1895, 1 and MRC is currently believed to be caused by a defect in closure of the median raphe during embryonic development 19 .…”

“…As far as squamomelanocytic tumors are concerned, the prognosis is worse, since these tumors have a more aggressive behavior similar to that of malignant melanoma. However, Pool SE refers no relapse or metastases for three patients with squamomelanocytic tumor for a mean follow up of 3.25 years [46,47].…”
