Pioglitazone, a PPARγ Agonist, Upregulates the Expression of Caveolin-1 and Catalase, Essential for Thyroid Cell Homeostasis: A Clue to the Pathogenesis of Hashimoto's Thyroiditis

Abstract: PPARγ may play a central role in normal thyroid physiology by upregulating Cav-1, essential for the organization of the thyroxisome and extracellular iodination. By upregulating catalase, PPARγ may also contribute to cell homeostasis. The inhibitory effect of Th1 cytokines on PPARγ expression may be considered as a new pathogenetic mechanism for HT, and the use of PPARγ agonists could open a new therapeutic approach.

“…Nthy-ori 3-1 cells, an immortalized thyroid follicular epithelial cell line, are highly suitable for studies of the control of growth and function in the human thyroid [37,38]. Our results are consistent with previous reports that a combination of IL-1α and IFN-γ induces Cav-1 reduction in primary cultures of human thyrocytes [18,39]. Collectively, these results confirm that Th1 cytokines are at least in part responsible for Cav-1 down-regulation.…”

“…12 Caveolin-1 protein expression on the apical membrane of thyroid follicular cells may contribute to maintaining cell homeostasis. 39 Decreased caveolin-1 expression may result in thyroid hormone synthesizes in the cytoplasm of thyroid follicular cells rather than classical synthesis sites on the apical membranes. This may induce an aberrant oxidative stress status in thyrocytes.…”

“…Peroxisome proliferator-activated receptor gamma (PPARγ) demonstrates protective effect in osteoarthritis in vivo; 40 on the other hand, PPARγ and its agonists are the known positive regulators of Cav-1 in different types of cells. 41 , 42 Such interaction corresponds to a generally positive effect of Cav-1 induction in inflammatory conditions 43 and indicates that Cav-1 modulation can be a target in osteoarthritic pain.…”
