Pioneer Factor Improves CRISPR‐Based C‐To‐G and C‐To‐T Base Editing

Abstract: Base editing events in eukaryote require a compatible chromatin environment, but there is little research on how chromatin factors contribute to the editing efficiency or window. By engineering BEs (base editors) fused with various pioneer factors, the authors found that SOX2 substantially increased the editing efficiency for GBE and CBE. While SoxN‐GBE (SOX2‐NH3‐GBE) improved the editing efficiency at overall cytosines of the protospacer, SoxM‐GBE/CBE (SOX2‐Middle‐GBE/CBE) enabled the higher base editing at P… Show more

“…Base editing efficiency is known to vary depending on Cas9 binding efficiency as well as the local sequence and chromatin context surrounding the target base 29,41,42 , and thus we expected gRNAs to vary substantially in editing efficiency across target sites. To account for this variability, we synthesized and cloned each gRNA paired with a 32-nt reporter sequence comprising the genomic target sequence of that gRNA into lentiviral base editor vectors ( Fig.…”

“…For example, fusion of T7 RNA polymerase with different deaminases (cytidine and adenosine deaminase), substantially widens the mutational spectrum in mammalian cells (Cravens et al, 2021). In addition, engineering BEs fused with additional chromatin modulating peptides, such as pioneer factor SOX2 (SRY-box transcription factor 2), to initiate chromatin unfolding and stimulate transcription, could be a promising strategy to further increase base editing efficacy (Yang et al, 2022).…”
