Piretanide and Urinary Excretion of Ions in Rat. Effect of Indomethacin

Abstract: 1. Piretanide is a new loop diuretic resembling furosemide. The effects of this diuretic on various parameters of renal function and on the excretion of the major ions were studied on anaesthetized, hydrated rats. 2. Piretanide induced a marked diuresis and increased sodium, potassium, chloride and calcium excretion. Calcium excretion increased more than that of sodium. Urinary osmolality and water reabsorption decreased. 3. Inulin clearance and phosphate excretion were significantly altered by piretanide. 4. … Show more

“…In clearance studies conducted in normal Wistar rats, piretanide, when administered at 1.5 mgkg i.v. during a state of normal hydration, caused a brisk diuresis, which waned rapidly within 20 min following drug administration (clearance studies consisted of five 10-min periods) (15). Inulin clearance (C1J was not significantly altered; consequently, no change in GFR was noted.…”

“…Nguyen-Simmonet et al (15) studied the effects of piretanide in rats using clearance techniques in the presence and absence of indomethacin. Figure 5 illustrates the effects of this combination on several excretion parameters.…”
