Pituitary adenylate cyclase activating peptide-27 – like immunoreactive nerve fibers in the mucosal layer of canine gastrointestinal tract in physiology and during inflammatory bowel disease

Abstract: Changes in the density of mucosal pituitary adenylate cyclase activating peptide-27 -like immunoreactive (PACAP-27 -LI) nerve fibers within various parts of the canine gastrointestinal (GI) tract during inflammatory bowel disease (IBD) were investigated. The distribution of nerves were studied, using a single-labelling immunofluorescence technique, in the mucosal layer of canine stomach, duodenum, jejunum, and descending colon. Canine IBD caused an increase in the density of PACAP-27-LI mucosal nerves in all s… Show more

“…The fragments of the colonic mucosal layer were then frozen at -20 8C, cut to 10-mm sections with a Microm cryostat (HM525, Walldorf, Germany) and mounted on microscopic slides. The sections were examined by the single immunofluorescence technique according to Gonkowski et al (2013). Briefly, the procedure was as follows: (a) drying for 45 min at room temperature (rt), (b) incubation with blocking solution (10% goat serum, 0.1% bovine serum albumin, 0.01% NaN 3 , Triton X-100 and thiomersal in PBS) for 1 h to avoid non-specific binding of antibody, (c) incubation with anti-SP antibody (rat, Biogenesis Ltd, Poole, UK, catalogue no.…”

“…The above-mentioned signs are accompanied by histopathological changes in the intestinal wall. In recent times, attention has been focused on the participation of intestinal innervation in pathological processes connected with IBD (Gonkowski et al, 2013;Rychlik et al, 2017;Brinkman et al, 2019), but the roles of particular neuronal active substances in the development of this disease are still not clear.…”

“…Results are presented as means ± standard error of mean (SEM). Nerve fiber distribution was assessed under × 40 objective by subjective observation (two independent researchers) as previously described (Gonkowski et al 2013 ) and depending on the density of labeling, described as not found or very few, small, moderate, and large number of nerve fibers.…”

“…The specimens were transferred to 18% phosphate-buffered sucrose solution with a temperature of 4 °C. Frozen sections of 10 µm were cut using a Microm cryostat (HM525, Walldorf, Germany), placed on gelatinised microscopic slides and subjected to single-cell immunofluorescence staining in accordance with the method reported by Gonkowski et al (2013b). The specimens were dried for 45 min at room temperature and rinsed three times in buffered NaCl solution (PBS, 0.1 mol, pH 7.4) for 15 min.…”
