Placental alpha microglobulin-1 (AmniSure® test) for detection of premature rupture of fetal membranes

Abstract: Detection of the PAMG-1 in the vaginal fluid using AmniSure(®) test is an accurate method to diagnose rupture of the fetal membranes, with high sensitivity, specificity, negative and positive predictive values.

“…7,11 Gestational age of the studied women calculated accurately from last menstrual period and early ultrasound scan done before 20 weeks. Examination of studied women followed by transabdominal ultrasound and vaginal speculum examination to visualize amnion leaking and for collection of samples for fern, nitrazine and AmnioQuick ® Duo + tests on admission.…”

“…False positive results are high with conventional diagnostic tests (fern and nitrazine) used to diagnose premature fetal membranes rupture. 7 AmnioQuick ® Duo + can detect amniotic fluid in a sample of vaginal secretions through identification of IGFBP-1 and AFP whose concentration in amnion is very high. 4,21 IGFBP-1 level in the amnion is 100-1000 times higher than in the serum.…”

“…Its concentration in amniotic fluid is greater than 1000-fold that found in normal vaginal secretions or maternal blood [6]. Recently, a rapid slide test that uses immunochromatography methods to detect trace amounts of PAMG-1 in cervicovaginal fluid has been developed to detect the rupture of membranes; the sensitivity of this method ranged from 94.4 to 98.9%, while the specificity ranged from 87.5 to 100% [6, 7]. …”
