Plant regeneration in Arachis stenosperma Krapov. and W. C. Gregory from roots and calluses derived from leaflets of in vitro plants

Abstract: Seed explants of A. stenosperma were cultured on MS medium supplemented with 6-benzylaminopurine with the aim of rescuing nonviable accessions stored in seed bank conditions. The regeneration potential of leaf explants from in vitro plants derived from embryonic axes was studied by using whole leaflets and leaflet segments. Explants were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of 6-benzylaminopurine and naphthalene acetic acid. Indirect organogenesis was observed … Show more

“…A maximum of 5.6 shoots was reported in A. stenosperma till date (Laxmi and Giri 2003a, b;Pacheco et al 2008). Rani and Reddy (1996) reported sporadic regeneration in A. duranensis and A. cardenasii; whereas a mean of 9.3 shoots per explant was obtained from A. cardenasii in this study.…”

“…A good regeneration protocol is an essential prerequisite for conservation as well as genetic manipulation. Earlier, explants like protoplasts, petiole, cotyledon, embryo axis, stem base callus and leaves (Li et al 1993;Rani and Reddy 1996;Gagliardi et al 2000;Laxmi and Giri 2003b;Mroginski et al 2004;Vidoz et al 2006;Pacheco et al 2008) were used to achieve morphogenesis in vitro with limited success in wild species of Arachis. Cultivated peanut appears to be an easy system for in vitro culture.…”

“…Gagliardi et al (2000) and Pacheco et al (2007aPacheco et al ( , 2008a described the production of single and multiple shoots from embryonic axes of several accessions of different sections through meristematic development in response to 8.8 lM 6-benzylaminopurine (BAP). High regeneration frequencies and multiple shoot formation rates were obtained from almost all studied species.…”

“…While the age of the donor plant did not influence regeneration, the regenerative potential was affected by the original position of the explant, decreasing from top to bottom. McKently et al (1991) and Pacheco et al (2008a) reported the occurrence of indirect organogenesis from leaflets of A. glabrata and A. stenosperma, respectively, in response to combinations of NAA and BAP. On the other hand, when NAA was used as the sole growth regulator, leaflets of A. stenosperma showed direct rhizogenesis, with subsequent shoot formation from adventitious roots, thus resulting in an additional multiplication system that can be used for conservation (Pacheco et al 2008a).…”
