Plant Transformation via Pollen Tube-Mediated Gene Transfer

Abstract: Genetic transformation using foreign genes and the subsequent development of transgenic plants has been employed to develop enhanced elite germplasm. Although some skepticism exits regarding pollen tube-mediated gene transfer (PTT), reports demonstrating improved transformation efficiency with PTT systems are increasing and encouraging and the adoption of increasingly refined pollenmediated methodologies may lead to species-dependent improvements in breeding. Here, we highlight PTT technology as an alternative… Show more

“…Pollen tube mediated transformation is efficient and simple alternative of producing transgenic plants, evading the stipulations for tissue culture. The PTT method was first reported in cotton (Gossypium hirsutum L.) [130] and rice [131]. Two approaches were as a naked DNA or through Agrobacterium.…”

“…Pollen transformations by Agrobacterium and biolistic method bypasses tissue culture regeneration step but frequently results in pollen with poor viability [ 105 , 106 ]. Furthermore, pollen-tube conversions can lead to chimerism [ 107 ]. While pollen magnetoreception has made progress in addressing these issues [ 106 ], it is still limited to dicots [ 108 ].…”

“…Agrobacterium and biolistic transformation of pollen also bypasses regeneration but often results in pollen with lower viability ( Wang et al, 2008 ; Zhao et al, 2017 ). In addition, pollen-tube transformations may result in chimerism ( Ali A. et al, 2015 ). While pollen magnetofection has improved on these drawbacks ( Zhao et al, 2017 ), its application remains constrained to dicots ( Vejlupkova et al, 2020 ).…”
