Plaque Calcification

Abstract: Vascular calcification (VC) is strongly associated with all-cause mortality and is an independent predictor of cardiovascular events. Resulting from its complex, multifaceted nature, targeted treatments for VC have not yet been developed. Lipoproteins are well characterized in the pathogenesis of atherosclerotic plaques, leading to the development of plaque regressing therapeutics. Although their roles in plaque progression are well documented, their roles in VC, and calcification of a plaque, are not well und… Show more

“…Atherosclerotic calcification in an advanced lesion may increase risk of cardiovascular events [44,45]. SMCs play critical roles in mediating intimal calcification by undergoing differentiation to osteoblast-like cells via increasing expression of calcification regulators, RUXN2, and BMP2 [46,47]. RUNX2, a decisive factor and early marker of SMC osteogenic transition and calcification that can be induced by BMP2, increases the expression of multiple osteogenic genes including collagen I, osteopontin, bone sialoprotein, and osteocalcin [11,48].…”

“…Arteriosclerotic calcification, commonly regarded as a latestage complication of atherosclerotic plaques, is an active process under tight regulations and is an independent predictor of cardiovascular morbidity and mortality (80). Although calcification of atherosclerosis plaques used to be considered unregulated and a passive deposition of calcium in the arterial wall, it is now well-recognized as a complex pathophysiological phenomenon resembling intramembranous ossification (81). YAP can interact with various factors involved in osteoblast development and maturation, such as Runx2 (82), bone morphogenic protein-2 (BMP-2) (83), and β-catenin (84).…”
