Plasma cells in multiple myeloma express a natural killer cell- associated antigen: CD56 (NKH-1; Leu-19)

Abstract: Bone marrow samples from 55 patients with multiple myeloma (MM) and 23 patients with monoclonal gammopathy of undertermined significance (MGUS) were evaluated with a broad panel of monoclonal antibodies. Plasma cells from 78% (43/55) of patients with MM strongly expressed the natural killer cell antigen CD56 (NKH-1, Leu-19). Of the 23 patients with MGUS, none showed strong CD56 reactivity, although three had weak reactivity in less than 20% of plasma cells. Myeloma cells expressing CD56 did not coexpress the C… Show more

“…Broadly, the differentially expressed surface proteins were markers of cell signaling such as cytokine receptors or cellular adhesion markers. CD150 53 , CD56 54 , CD28 55 are known to be expressed on normal or malignant PC; however, CD102 and CD154 are not known to be expressed on LLPC. Surface protein expression of CD31 and CD49d were also observed on the BM-resident LLPC population, consistent with previous studies of BM-resident LLPC in NHP (Supplementary Data 4 ) 37 .…”

“…The neural cell adhesion molecule (NCAM), known as CD56, is a membrane glycoprotein and member of the immunoglobulin superfamily. CD56 is an NK-cell surface marker not expressed in normal plasma cells, but 78% of MM cells showed CD56 positivity [ 87 ]. To counter antigen escape, a multitarget CAR was developed, targeting CD56 among others (NCT03271632).…”

“…CD56 is expressed on neural cells, muscle tissues, and various lymphoid cells. It is expressed on a small percent of normal PC but overexpressed in about 65–80% of malignant PC dyscrasias, especially MM [ 73 , 74 , 75 ]. Overexpression of CD56 promotes the transcription of CREB1 targets, the anti-apoptotic genes BCL2 and MCL1, resulting in a robust anti-apoptotic effect [ 76 ].…”
