We tested the hypothesis that indirect measures of oxidative stress (vitamin E, glutathione peroxidase, and malondialdehyde) differ in dogs in heart failure resulting from either myxomatous mitral valve disease or dilated cardiomyopathy. Dogs were classified according to the International Small Animal Cardiac Health Council (ISACHC) classification. Additionally, the effect of cardiac therapy on oxidative stress parameters and N-terminal pro-B-type natriuretic peptide (NT-proBNP) in advanced stages of congestive heart failure was investigated. There were no significant differences in oxidative stress parameters between healthy dogs and the individual groups of cardiac patients. Significantly lower malondialdehyde (MDA) was observed in the ISACHC II group in comparison to ISACHC groups III and I. A significant positive correlation in treated patients was observed between NT-proBNP and MDA, NT-proBNP and vitamin E, as well as between MDA and vitamin E (and lipid-standardized vitamin E). No significant differences in any of the measured parameters were found between treated and non-treated cardiac patients. Our results suggest an association between MDA (the extent of lipid peroxidation) and NT-proBNP, vitamin E and NT-proBNP, as well as between MDA and vitamin E in treated canine patients. Plasma vitamin E concentration was maintained in all stages of cardiovascular disease in these canine patients.

Among species, larger animals tend to live longer than smaller ones, however, the opposite seems to be true for dogs—smaller dogs tend to live significantly longer than larger dogs across all breeds. We were interested in the mechanism that may allow for small breeds to age more slowly compared with large breeds in the context of cellular metabolism and oxidative stress. Primary dermal fibroblasts from small and large breed dogs were grown in culture. We measured basal oxygen consumption (OCR), proton leak, and glycolysis using a Seahorse XF96 oxygen flux analyzer. Additionally, we measured rates of reactive species (RS) production, reduced glutathione (GSH) content, mitochondrial content, lipid peroxidation (LPO) damage and DNA (8-OHdg) damage. Our data suggests that as dogs of both size classes age, proton leak is significantly higher in older dogs, regardless of size class. We found that all aspects of glycolysis were significantly higher in larger breeds compared with smaller breeds. We found significant differences between age classes in GSH concentration, and a negative correlation between DNA damage in puppies and mean breed lifespan. Interestingly, RS production showed no differences across size and age class. Thus, large breed dogs may have higher glycolytic rates, and DNA damage, suggesting a potential mechanism for their decreased lifespan compared with small breed dogs.

Purpose: Oxidative mechanisms play a major role in the etiology and pathogenesis of cataract, especially age-related cataract (ARC). Our study aimed to investigate the serum vitamin E level and the activities of systemic antioxidant enzymes: superoxide dismutase (SOD) and glutathione peroxidase (GPX) in the blood of dogs with age-related cataract but without any general diseases. Material and methods: The population examined consisted of 40 dogs of various breeds and both sexes, aged 10-16 years, diagnosed with age-related cataract. Cataract was diagnosed by complete ocular examination. Blood was collected during clinical examination from the external jugular vein in dogs weighing less than 20 kg or from the small saphenous vein in dogs weighing over 20 kg. The control group consisted of 10 healthy dogs of different breeds and sexes, aged 8-12 years, without cataract. SOD and GPX were measured by a colorimetric reagent kit Randox SOD and Randox GPX. The level of vitamin E was determined using a Perkin-Elmer LS 30 luminescent spectrometer. This apparatus determines the vitamin E level by the fluorometric method of Hansen and Warwick . Results: The results showed a significantly lower activity of SOD as well as a lower activity of GPX and a lower level of the vitamin E in dogs with ARC compared to healthy dogs. Conclusions: The results obtained indicate a weakening of the general antioxidant defence system in the course of ARC in dogs. These results suggest that such conditions may have effect on the development of canine age-related cataract and are consistent with the free radical theory of age-related cataract development.
