Plasmacytoid Dendritic Cells Induce CD8+ Regulatory T Cells In Human Ovarian Carcinoma

Abstract: To directly dissect the role of each immune component in human tumor immunopathogenesis, we have studied the interaction between dendritic cells and T cells in the tumor environment of patients with ovarian carcinoma. We previously reported that functional plasmacytoid dendritic cells, but not functionally mature myeloid dendritic cells, accumulated in tumor microenvironments. We now show that tumor ascites macrophage-derived dendritic cells induced tumor-associated antigen-specific CD8+ T cells with effector … Show more

“…Human pDCs are regarded as immunomodulating cells and have been described to induce immunity (31), as well as to promote regulatory T cell-mediated immunosuppression (32,33). In this study, we investigated whether pDCs can play a role in immune responses against exogenous particulates.…”

“…Activated pDCs have been demonstrated to promote Th1 (31,47) as well as Th2 responses (32,33), underlining their remarkable functional plasticity. The pDCs were shown to prime specific CD4 + and CD8 + lymphocytes against endogenous and exogenous Ags (15,48), viruses (49) and tumor Ags (45).…”

“…On the contrary, pDC may regulate anti-tumor immunity and support immune evasion and tumor escape. They exhibit reduced IFN- production upon TLR9 stimulation and can induce IL-10 producing CD4 + and CD8 + Treg [35,36]. This suggests that anti-tumor immune responses can be regulated through both modulation of pDC function by the Takata et al, 13 tumor and by limiting anti-tumor cytolytic activity through induction of CD8 + Treg.…”

“…Similarly, immature conventional DC can induce CD8 + Treg (17) and FoxP3 + alloantigen-specific CD8 + CD28 -T suppressor cells can also be generated in vitro by tolerized endothelial cells (18). Primary suppressive CCR7 + CD45RO + CD8 + T cells have been isolated from human tumors, suggesting that they may contribute to the tumor-induced immunosuppressive network (19).…”
