Plasmolysis ofEscherichia coliB/r with Sucrose

Abstract: Escherichia coli B/r cells were plasmolyzed in sucrose solutions and observed under phase contrast. The prevalence of plasmolysis under various conditions was noted, and the degree of plasmolysis was categorized as slight, extensive, or severe. The presence of ions reduced the prevalence of plasmolysis. Survival curves showed that extensive plasmolysis was not lethal to colony-forming ability. Plasmolysis represents a dramatic morpho

“…s~ NaC1) in the medium, then the cells frequently have a distorted appearance: they collapse into irregular flat, elongated discs, which, according to their orientation with respect to the axis of observation, appear either similar or unaltered, or as widened cells or distorted, relatively thin rods (Fig. 6), confirming previous reports (Schleie & Dalen, 1968;Schleie, 1969). In contrast to the plasmolytic bays, produced by weaker shocks, these results are difficult to achieve reproducibly.…”

“…In a further analysis of the consequences of pnd gene expression we have found a drastic effect on the response of cells to hypertonic medium. Ordinarily, E. coli cells are plasmolyzed in such a medium: internal water is lost, the cytoplasm shrinks, and the cytoplasmic membrane is caused to retract from the cell wall (3,8,32). But when we exposed the cells to a hypertonic sucrose solution after the addition of rifampicin to induce the pnd protein, plasmolysis was largely abolished.…”
