Platelet Aggregation Is Not Initiated by Platelet Shape Change

Abstract: SUMMARY:Because the initial decrease in light transmission in platelet aggregometry is attributed to platelet shape change, it is widely held that platelet shape change is a prerequisite for platelet aggregation. We conducted this study to determine the basis of this initial optical effect in aggregometry. Platelets were activated with ADP, thrombin, or the thrombin receptor agonist peptide SFLLRN (TRAP 1-6 ). In every case the initial decrease in light transmission occurred with the concomitant formation of m… Show more

“…Our technique of assessing PSC is highly sensitive (resolution 0.07 fL) 18,20,22 when compared with others who used less sensitive methods. 23,24 We found that fib (0.5-2 g/L) had no effect on PSC at various time points (between 1 and 20 minutes), which was in agreement with the findings of others. 25 Also, ADP-induced and 5HT-induced PSC was not inhibited by tirofiban (50-250 ng/mL), a potent and specific GP IIb/IIIa receptor antagonist.…”

“…It has been shown that even fibrinogen-coated latex spheres form tight aggregates in vitro after the addition of thrombin [119]. This finding might suggest that serotonin is less important for aggregate formation than for aggregate stability and the prevention of premature clot lysis.…”

“…The aggregometric profile also reflects this distinction. The earlier notion, that initial decrease in light transmission in platelet aggregometry attributed to platelet shape change, which was known to be a prerequisite for platelet aggregation, has been questioned [12]. The initial decrease in light transmission is now believed to be the outcome of microaggregate formation.…”
