Platform- and label-free detection of lead ions in environmental and laboratory samples using G-quadraplex probes by circular dichroism spectroscopy

Abstract: Guanine-rich quadruplex (G-QD) are formed by conversion of nucleotides with specific sequences by stabilization of positively charged K+ or Na+. These G-QD structures differentially absorb two-directional (right- and left-handed) circularly polarized light, which can discriminate the parallel or anti-parallel structures of G-QDs. In this study, G-QDs stabilized by Pb2+ were analyzed by a circular dichroism (CD) spectroscopy to determine Pb2+ concentration in water samples. Thrombin aptamer (TBA), PS2.M, human … Show more

“…A more pressing concern is the ability to distinguish Pb 2+ from K + because natural water samples typically contain appreciable quantities of potassium, which can potentially suppress a probe response for the presence of lead and generate a false-negative readout . Recently, Kim and co-workers utilized CD to determine a K d of 830 nM for Pb 2+ -binding by native TBA in the presence of 100 μM K + ion, which increased to 220 μM in the presence of 100 mM K + . Native TBA binds K + ion with a K d of 5000 nM (from mass spectrometry)13 000 nM (from CD titration) depending on the conditions.…”

“…A lead ion also more strongly stabilizes H-bonds through polarization enhancement (Figure B), with a diagnostic long-wavelength circular dichroism (CD) maximum at ∼310 nm rather than ∼290 nm for monocationic antiparallel GQs . This feature has been employed by Kim and co-workers to monitor Pb 2+ -binding using the 15mer thrombin binding aptamer (TBA, 5′-G 2 T 2 G 3 TGTG 2 T 2 G 2 -3′) as a CD-specific label-free sensor …”

“…Other sources include lead battery manufacturers, lead-based pigment manufacturers, automobile industries, decayed water-carrying metal pipes and contaminated consumer products [ 20 ]. Exposure to lead at a certain concentration has been implicated with several health problems such as renal dysfunction, nervous system impairment, headache, fatigue, constipation and brain development impairment in children [ 21 , 22 , 23 , 24 , 25 , 26 , 27 , 28 ].…”

“…Circular dichroism (CD) is very sensitive to secondary structure of nucleic acid [ 64 ]. The spectral shape and signal of CD can effectively show the secondary structure changes of DNA caused by the binding to small molecules [ 65 ]. Isothermal titration calorimetry (ITC) is an intuitive and unlabeled method for analyzing and characterizing molecules which can directly measure the thermal changes caused by the chemical reactions between the two components [ 66 , 67 ].…”
