Pleiotropic Effects of IGF1 on the Oocyte

Abstract: A woman’s endocrine system plays a crucial role in orchestrating cellular interactions throughout her life. The growth hormone (GH) and insulin-like growth factor (IGF) system appears to impact crucial reproductive events and cell types of the ovary, such as granulosa cells, theca cells, and oocytes. Further, IGF1 is a cornerstone during embryonic development and influences predominantly developing and pre-antral follicles. In this commentary, we will emphasize the pleiotropic effects of IGF1 on physiological … Show more

“…Unlike FGF2, LIF acts through two signaling cascades, MAPK3/1 and the signal transducer and activator of transcription 3 (STAT3)-dependent pathways, but is more heavily controlled by JAK/STAT3 [ 8 ]. Similarly, IGF1 acts on both the oocyte as well as the embryo, increasing oocyte quality and embryonic development through the phosphoinositide 3-kinase (PI3K)/protein kinase B (AKT)/mammalian target of rapamycin (mTOR) pathway [ 9 , 10 ].…”

“…Each cytokine has beneficial effects on in vitro maturation that are amplified when the cytokines are added in combination. FGF2 increases cumulus cell expansion [ 7 ], LIF increases oocyte maturation and early embryonic development [ 8 ], and IGF1 increases oocyte quality and embryonic development [ 9 , 10 ]. Researchers investigated the effects of maturation medium supplementation with these three cytokines, termed FLI, in the porcine model.…”

“…The vast majority of PGCs and primary oocytes undergo caspase-dependent apoptosis [ 68 , 69 ]. The survival and proliferation of PGCs are crucially dependent on external signaling molecules such as KIT ligand (KITLG (SCF)), Leukemia inhibitory factor (LIF), Insulin like growth factor 1 (IGF1), retinoic acid, and cAMP activators [ 70 , 71 ]. The presence of KITLG and LIF is essential to prevent PGC apoptosis in vitro [ 60 ], and both factors are secreted by pre-granulosa and granulosa cells, promoting PGC survival in vivo [ 61 , 62 ].…”
