Pleural Fluid Transforming Growth Factor–β1 Correlates with Pleural Fibrosis in Experimental Empyema

Abstract: Transforming growth factor-beta1 (TGF-beta1) is a growth factor that is implicated in fibrosis of many organs. The purpose of this study was to determine the sequential levels of TGF-beta1 in the pleural fluid of rabbits that had undergone empyema induction, as fibrosis of the pleural space develops. Thirty-seven rabbits underwent empyema induction. Rabbits were sacrificed on Days 1, 2, 3, 4, 5, 6, and 8. Pleural fluid and viscera pleura specimens were collected at autopsy. TGF-beta1 levels were measured in pl… Show more

“…Anti-TGF-␤ strategies are a promising therapeutic approach for prevention or cure these complications. One study has shown that intrapleural injection of TGF-␤ Abs can reduce empyema-induced pleural fibrosis in a rabbit model (11,12). Although the role of TGF-␤ in other pleural fibrotic disorders is not definitively proven, we believe that our model could be very useful to further understand how to halt the progression or even cure this chronic condition.…”

“…It has recently been shown that intrapleural injection of TGF-␤2 stimulates mesothelial cells to produce collagen and endogenous TGF-␤, further increasing the production of matrix proteins (10). Human mesothelial cells are not only able to synthesize TGF-␤ but also have receptors for this cytokine (11,29), further supporting the hypothesis of autocrine activation of TGF-␤ in pleural fibrosis.…”

“…Secretion of TGF-␤ and IL-10 by DOC, MSC, DOC-IFN-␥, and MSC-IFN-␥ were tested with the medium collected from the wells of MLC plates. The assay procedure followed instructions of the BioSource Mouse IL-10 ELISA kit (BioSource International) (36) and the Quantikine TGF-␤1 Immunoassay kit (R&D Systems) (37,38). Absorbance was measured with an ELx800UV ELISA plate reader at 450 nm.…”

“…3) The final ''organising'' stage of pleural infection is characterised by fibroblast growth and proliferation, resulting in the formation of an inelastic, fibrous pleural peel which impairs lung re-expansion. The mechanisms which underlie the development of pleural fibrosis in empyema are poorly understood, although recent work using a rabbit model of empyema has suggested an important role for the fibrogenic transforming growth factor-b1 [18].…”
