Polarimetric Research of Pharmaceutical Substances in Aqueous Solutions With Different Water Isotopologues Ratio

Abstract: Objective: Methodology development for quality control of optically active pharmaceutical substances based on water isotopologues. Methods: Solutions of L-ascorbic acid, glucose, galactose and valine stereoisomers were prepared using deuterium depleted water (DDW-«light» water, D/H=4 ppm), natural deionized high-ohmic water (BD, D/H=140 ppm), heavy water (99.9% D2O). The optical rotation was observed using an automatic polarimeter Atago POL-1/2. The size distribution of giant heterogeneous clusters (GHC) of wat… Show more

“…But any complication of the system due to the addition of excipients to the aqueous solution leads to a decrease in lifetime leads of the biological object. At the same time, there is also a noticeable increase in the specific optical rotation, which reflects a change in the chiral properties of the API solution [1,2]. ; (pH=6,30, n=3, P=0,95).…”

“…Since the middle of the last century, the stability of ascorbic acid (AA), which has antioxidant properties in mammals and in foodstuffs, has been actively studied [1][2][3][4][5][6][7][8][9]. AA degradation can occur spontaneously, even in an anaerobic environment, and under the influence of a number of factors [1,2,5]. An increase in temperature and in alkalinity of its solution, exposure to radiation, the presence of d-elements ions and compounds with a more positive redox potential are the main causes of AА degradation.…”

“…Due to the polarity of DMF molecules, it was of interest to study the initiation of optical activity in solvent associates under the influence of chiral substances, as was found for giant heterogeneous water clusters [9][10][11][12]. It turned out that in the region of low concentrations (≤0.1%), the specific optical activity of L-ascorbic acid solutions in DMF drops sharply ( fig.…”

“…Accordingly, the biological activity of solutions of the dispersed lactose monohydrate samples exceeds that of the native samples, which may be associated with the mechanochemical activation [19]. The highest biological activity corresponds to the solutions of the dried lactose monohydrate samples.…”
