The electrochemical behavior of two novel triazolobenzodiazepines (adinazolam and U43,465F) has been studied in the pH range 3 to 13.5. Both compounds are reduced on the dropping mercur). electrode and yield one reduction wave attributed to the azomethine group. The two-electron transfer is polarographically reversible. However, the transfer is followed by a fast conversion of the electrolysis product by a chemical reaction with an irreversible global process. A weak adsorption of the reactant has been observed under certain conditions. Quantitative analysis has been performed, showing that adinazolam and U43,465F can be determined up to 1.1 X polarograp hy.KEY WORDS: Triazolobenzodiazepine, Voltammetn. M using differential pulse

Apparent kinetic constants of the reversible opening of a triazolo-1,4-thienodiazepine were determined by UV-spectrophotometry and by polarography in water at several pH values assuming as a hypothesis a stationary state for the carbinolamine intermediate. Both the apparent kinetic constants, k f and k r , exhibited a maximum for the values H 0 ϭ Ϫ0.25 and pH ϭ 6.07. A possible determination of the elementary kinetic constants of the several acidobasic species which may be involved in the opening and closing process according to the pH and pKa values was studied and is discussed. The results are consistent with the hypothesis that both the opening of the diazepine cycle and the closing of the opened form proceed through a mechanism suggesting that the protonated form of the carbinolamine function of the intermediate is involved.

The pK a values of the freely water-soluble opened form of a thieno-1,2,4-triazolo-1,4-diazepine, namely 2-(3-aminomethyl-5-methyl-1,2,4-triazolo)-3-(2-chlorobenzoyl)-[2,3-c]thienopiperidine, were determined. pH measurements were made in the range 1 < pH < 10. Two of the three pK a values found in this range significantly overlapped; they quantify the ionization of both the aminomethyl residue and the piperidino moiety of the molecule. The third corresponds exclusively to the ionization of the triazolo nucleus. The pK a value of the thienophenone was obtained by UV spectrophotometry using acidity functions. All results were extracted from experimental data by a curve-fitting method.
