Pollen Fertility Assessment Through Acetocarmine Staining and in Vitro Germination in Solanum Tuberosum L.

Abstract: The wide occurrence of potato genotypes characterized by male sterility considerably complicates the realization of breeding programs. Therefore, male fertility in potato germplasm requires assessment as an important task. In the fresh study, 57 breeding lines from the Russian Potato Research Center and 85 cultivars of different origins underwent acetocarmine staining and in vitro germination assessment. With acetocarmine staining, 22 samples exhibited with tetrad sterility. In the remaining 120 samples, stain… Show more

“…Pollen germination indicated pollen viability, while pollen tube length showed vigor (Sulusoglu and Cavusoglu, 2014). Pollen germination is one of the methods used to assess pollen fertility, previously carried out on 142 accessions of Solanum tuberosum L. (Gins et al, 2022). For male dioecious flowers of V. pubescens, the pollen germination was faster than in monoecious plants, indicating the need for male plants in the cultivation.…”

“…Sesbania is a nitrogen-fixing shrub that can restore disturbed soils by improving soil nitrogen (N2) and fertility via the nodulation process between bacterial nodules and sisban roots and can also serve as fodder and wood fuel (Grzeça et al, 2021). Sesban species propagation is often by the seeds and represents natural resources for reproduction and maintenance of genetic diversity in crop plants (Taiz and Zeiger, 2002;Gins et al, 2022;Narimonov et al, 2023).…”

“…The importance of sweet potato as a food crop flourishes rapidly in some parts of the world. It is also a good food-security crop and sells for cash by poor resource farmers (Tairo and Kullaya, 2004;Dalamu et al, 2012;Rawaida and Nur-Fazliana, 2019;Haq et al, 2021;Gins et al, 2022).…”
