Poly(2‐(dimethylamino) ethyl methacrylate)‐b‐poly(lauryl methacrylate)‐b‐poly(oligo ethylene glycol methacrylate) triblock terpolymer micelles as drug delivery carriers for curcumin

Abstract: This work aims to investigate the ability of poly[2‐(dimethylamino)ethyl methacrylate‐b‐lauryl methacrylate‐b‐(oligo ethylene glycol)methacrylate], PDMAEMA‐b‐PLMA‐b‐POEGMA amphiphilic triblock terpolymers, to encapsulate the model hydrophobic compound curcumin. The terpolymers were synthesized by RAFT polymerization methodologies at variable molecular weights and compositions and self‐assemble into spherical micelles with mixed PDMAEMA/POEGMA coronas and PLMA cores, when inserted in aqueous solutions. “…It is interesting to note that Cur-TA-MP2 showed an overall higher DL % and EE % compared to Cur-MP1, which may suggest a stronger drug–polymer interaction due to additional hydrophobicity in the core environment of Cur-TA-MP2. Cur-TA-MP2 had EE % of >80%, which was significantly higher than the Cur-loaded micelles from a linear triblock copolymer, poly[2-(dimethylamino)ethyl methacrylate- b -lauryl methacrylate- b -(oligo ethylene glycol)methacrylate] of about 12% …”

“…Poly (2‐(dimethylamino) ethyl methacrylate) (pDMAEMA) is a water‐soluble cationic polymer, which can electrostatically interact with DNA and form a stable polyelectrolyte complex. [ 145 ] The transfection efficiency of pDMAEMA was improved with the increase of polymer molecular weight. Since the average pKa of amino group is 7.5, the tertiary amino group of pDMAEMA has been partially protonated in physiological solution, so it can have certain proton buffering in the acid endosomal compartment.…”

“…The current study investigates the synthesis of a series of homopolymeric and copolymeric injectable hydrogels based on PEGMEM and DMAEM for their use in cartilage regeneration. Both monomers have been reported to be used for several biomedical applications [ 27 , 28 , 29 , 30 ]; however, the safety of polymers based on DMAEM is still controversial [ 31 , 32 , 33 ]. The previously described synthesis procedure [ 26 ] was modified to obtain more flowable hydrogel to fulfill the injectability requirement.…”
