Poly(I:C) adjuvant strongly enhances parasite-inhibitory antibodies and Th1 response against Plasmodium falciparum merozoite surface protein-1 (42-kDa fragment) in BALB/c mice

Abstract: Malaria vaccine development has been confronted with various challenges such as poor immunogenicity of malaria vaccine candidate antigens, which is considered as the main challenge. However, this problem can be managed using appropriate formulations of antigens and adjuvants. Poly(I:C) is a potent Th1 inducer and a human compatible adjuvant capable of stimulating both B- and T-cell immunity. Plasmodium falciparum merozoite surface protein 1 (PfMSP-1) is a promising vaccine candidate for blood stage of malaria … Show more

“…Interestingly, the ratio of IgG2a/IgG1 showed no significant difference between SBLP alone and with an adjuvant. However, in most studies, ISA 201VG and Poly (I:C) adjuvants induced a Th1-polarized immune response [50,51]. We assumed that the presence of the GEM particles may change this bias.…”

“…The avidity of anti-rPoMSP-8 IgG antibodies was estimated as described previously [22]. Briefly, ELISA testing was performed as described above, except that the test was performed in duplicate plates.…”

“…[ 225 , 226 ] For example, as an adjuvant, alum normally generates IgG1 as the dominant subclass with very minor IgG2 a,c levels, while Th1‐supporting adjuvants, poly I:C and CPG ODN, generate IgG2 a,c as the dominant subclass. [ 227 , 228 ] Recently, SARS‐2‐S protein was tested for its efficacy in generating nAbs against SARS‐2 in combination with adjuvants, such as AS03, AS37, Alum, and Alum‐CPG1018. AS03 followed by Alum‐CPG1018 outperformed the other adjuvants tested in terms of generating stable, high levels of nAb titers > 10 3 ( n = 5–6 mice per group).…”
