Polyamine Metabolism in P388 Leukemia Cells and in Ascites Tumor-Bearing Mice

Abstract: Alterations in ornithine decarboxylase (ODC) activity and in the polyamine and nucleic acid (RNA and DNA) concentrations were studied in P388 leukemia cells and in the host liver at different times after intraperitoneal inoculation of 5 × 106 tumor cells into BDF1 mice. ODC activity of leukemia cells had a maximum at day 5 after tumor transplantation. Cellular spermidine concentration declined significantly during the first 5 to 6 days. The RNA concentration of tumor cells changed paralle… Show more

“…We have shown earlier that P388/S leuke mia cells grown intrapcritoneally in BDF| mice have an active polyamine metabolism at the early phase or cell proliferation (1-2 days), but a continuous decrease in all poly amine fractions can be observed throughout tumor growth [19,23]. Our present results indicate (table I) that ODC activities of P388/S leukemia cells originating from DFMO-treated mice were very low and just detectable during the whole period of experi- merits.…”

“…DFMO hydrochloride monohydrate (RMI 71.782, DFMO kindly donated by Centre de Recherche Merrell International, Stras bourg, France) was dissolved every 2 days in distilled water at a concentration of 2% for administration via drinking water. P388/S leukemia cells were main tained in BDF| mice by serial transplantation in our Institute as previously described [19]-DFMO was given to BDF| mice, 60-70 in each group, in water as sole drinking fluid. 2 weeks before the intraperitoneal transplantation of P388/S leuke mia cells and continued throughout tumor growth.…”

“…For vacuumpackaged meat stored at low temperatures, Enterobacteriaceae, especially Hafnia alvei and Serratia liquefaciens, have been identified as playing a major role in the accumulation of diamines (Dainty et al, 1986). The situation is complicated by the fact that socalled polyamines constitute essential compounds of living cells (Kremmer et al, 1984), while other amines cause adverse health effects (Bardocz, 1995) such as histamine (reviewed by Lehane and Olley, 2000) and tyramine (Beutling, 1996). Other amines potentiate such effects or are associated with tumorous growth (spermine, spermidine and putrescine; Kaminski et al, 1987;Seiler, 2003).…”

“…(1) P388 cells in culture represent a poly amine pattern similar to those growing in vivo [8], characterized by the predominance of Spd, while Pu and Spn contributed to the total polyamines in a smaller extent only.…”
