Polycomb recruitment to DNA in vivo by the YY1 REPO domain

Abstract: Polycomb group (PcG) proteins are responsible for maintaining transcriptional repression of developmentally important genes. However, the mechanism of PcG recruitment to specific DNA sequences is poorly understood. Transcription factor YY1 is one of the few PcG proteins with sequence-specific DNA binding activity. We previously showed that YY1 can recruit other PcG proteins to DNA, leading to histone posttranslational modifications and stable transcriptional repression. Using Drosophila transgenic approaches, … Show more

“…Recent evidence suggests that other changes in metabolism and adult physiology might also contribute to the increased longevity of PRC2 mutants. YY1, the mammalian homolog of Drosophila PHO (a DNA-binding PcG protein involved in recruiting PRC2 to chromatin), directly regulates many genes required for mitochondrial oxidative metabolism (51)(52)(53). It will be interesting to determine whether transcriptional regulation of metabolic genes is a broader theme in the adult function of PcG proteins.…”

“…The N terminus has been implicated in transcriptional activation, whereas residues 170 -201 participate in transcriptional repression (12,13). Residues 201-226 form the REPO (recruitment of polycomb) domain, involved in polycomb group repression (14).…”

“…35 Recent reports suggest that YY1, the mammalian ortholog of the Drosophila PRE DNAbinding protein PHO and RYBP, which interacts with both PcG proteins, is involved in the PRC2 recruitment process. [43][44][45] Long non-coding RNAs (ncRNA) have also emerged as potential elements involved in PRC2 recruitment. HOTAIR ncRNA promote PRC2 recruitment in trans and is associated with the transcriptional repression of the HOXD locus.…”
