“…It has been reported that the healing process of bone tissue begins with inflammation following immune cells, releasing inflammatory factors at the initial stage, consequently recruiting MSCs and guiding their osteogenic differentiation. This process is also closely related to neovascularization [ 55 ]. Therefore, the modulation of local immune responses following implantation is of great importance for bone regeneration.…”

“…The sequence and duration of their appearance often affect the result of bone repair. Excessive immune activation caused by bacteria and chronic diseases can lead to long-term M1 polarization and chronic inflammation, therefore, reducing M1 polarization and inducing M2 polarization to remodel the local microenvironment is beneficial to bone regeneration. , However, in the case of trauma or resection, the transient appearance of M1 macrophages after biomaterial implantation will produce the necessary inflammatory response, inducing a transient M1 polarization of macrophages and then interacting with BMSCs belonging to the skeletal system to guide its phenotypic changes to M2 in time, which seems to be a way closer to physiological bone healing . In our current research, the LL37 peptide mainly induces pro-inflammatory response in the early stage, and is more inclined to activate the M1 polarization of macrophages.…”

“…As shown in Figure 11 , an increase in Cav1.3 induces an increase in the cytosolic Ca 2+ concentration of H9c2 cells, thereby increasing the activation of Ca 2+ -dependent signalling, Rcan1, calcineurin, and calpain, which subsequently upregulates the gene expression of hypertrophy markers and upregulates the gene expression of inflammatory cytokines by activating the NF-κB/p65-dependent signalling pathway [ 21 , 22 , 23 , 24 , 25 , 26 , 27 , 28 ], causing cardiac hypertrophy and inflammation. Meanwhile, the elevation of Gal-3, RhoA, and Rock further increased ROS [ 27 , 28 , 29 , 30 , 31 , 32 ], subsequently regulating the Bcl-2/caspase-3-dependent apoptotic signalling pathway [ 33 , 34 ], thereby leading to cell rupture and fibrosis.…”
