Polymeric carrier-mediated intracellular delivery of phosphatidylinositol-3,4,5-trisphosphate to overcome insulin resistance

Abstract: Inducing insulin actions by intracellular PIP3 delivery (PEI-25/PIP3 complexes) in some forms of insulin-resistant cells provides the first proof-of-principle for the potential therapeutic use of PIP3 in a "second-messenger agonist" approach. In addition, utilization of an artificial bio-mimetic membrane platform to screen for highly efficient PIP3 delivery predicts biological function in cells.

“…The outcomes of this experiment are similar to those of an earlier study employing PEI25 as a carrier for PIP3 [51]. Polyethyleneimine (PEI-25, 25 kDa) was previously identified as an effective vehicle for PIP3, forming complexes observed on the cell membrane within approximately one minute.…”

“…Mice with deficient PI3K subunit (heterozygous for p110α or p110β) displayed impaired glucose metabolism (Figure ) . In addition, exogenous PIP3 supplementation in L6 muscle cells increased both glucose uptake and glucose utilization and the effect was mediated by the activation of GLUT4 …”

“…63,64 In addition, exogenous PIP3 supplementation in L6 muscle cells increased both glucose uptake and glucose utilization and the effect was mediated by the activation of GLUT4. 65 PIP2 also participates in the insulin secretion process. As plasma glucose rises, the ATP concentration increases and ADP concentration decreases, resulting in K(ATP) channel closure, membrane depolarization, Ca 2+ influx and insulin release ( Figure 3).…”

“…The researchers condensed polyethyleneimine (PEI) 25 kDa and PI(3,4,5)P3 into PEI-25/PIP3 nanoparticles whose cationic charge (derived from PEI25) caused PI(3,4,5)P3 to be retained at the cell perimeter. This strategy was thought to mimic the extent and cellular organization of PI(3,4,5)P3, which is preferable for insulin stimulation ( Kachko et al ., 2015 ).…”
