Polymorphisms of COMTVal158Metand DAT13′-UTR VNTRin Illicit Drug Use and Drug-Related Psychiatric Disorders

Abstract: To investigate the involvement of COMT Val158Met and DAT1 3'-UTR VNTR genotypes in the pathogenesis of illicit drug use and drug-induced psychotic disorders (DIP), 187 substance users and 386 normal controls were recruited from Northern Taiwan. Substance users and normal controls significantly differed in allele frequencies of COMT Val158Met (p = 0.039) but not in allele frequencies of DAT1 3'-UTR VNTR (p = 0.879). However, neither allele frequencies of COMT Val158Met nor allele frequencies of DAT1 3'-UTR VNTR… Show more

“…According to Volkow et al, dopamine plays a critical role in the treatment of addictive disorders, 4 and Le Foll et al proposed that a genetic study should evaluate this role 5 . Moreover, the catechol‐O‐methyltransferase (COMT) polymorphism considerably affects the treatment of substance and behavioral addictions 6,7 . Nonetheless, its association with IGD needs further evaluation.…”

“…Compared with the Val allele, the Met allele is more associated with lower COMT enzyme activity, lower dopamine degradation, and higher synaptic dopamine 8 ; consequently, the Met allele is associated with more efficient frontal lobe functions 9 . Catechol compounds and the frontal lobes, associated with cognition, have major roles in various psychiatric disorders, including addictive disorders 6,7 …”

“…Since dopamine is the main neurotransmitter involved in the reward pathway, genes related with dopamine synthesis, degradation, receptors, and transporters are possible research topics [13]. In this context, the COMT Val 158 Met (rs4680) polymorphism has been studied in the scope of drug addiction with evidence of a signi cant correlation in drug addicted patients [14,15]. The COMT Val 158 Met polymorphism determines the dopamine concentration in the prefrontal cortex (PFC), but not in the striatum [16].…”

“…Differences in catechol-o-methyl transferase (COMT) genotype have been linked to illicit drug use (Chen et al, 2014) and polysubstance abuse (Vandenbergh et al, 1997). However, with regard to alcohol use, the role of COMT is unclear.…”
