Polymorphisms of TNF microsatellite marker a and HLA-DR-DQ in diabetes mellitus—a study in 609 Swedish subjects

Abstract: ABSTRACT:We explored the importance of the genetic markers; microsatellite TNFa, HLA-DR3-DQ2 and DR4-DQ8 in diabetes mellitus. The studied groups were as follows; autoimmune type 1 (n=63), non-autoimmune type 1 (n=35), latent autoimmune diabetes in adults (LADA; n=54) and non-autoimmune type 2 (n=340) and these patients were compared to 117 healthy controls. HLA genotyping was done with polymerase chain reaction and sequence-specific oligonucleotides. TNFa microsatellites were determined with polymerase chain … Show more

“…The most common TNFa microsatellite in the European population was TNFa2 with a frequency varying between 20.8% in the French sample and 32% in a Danish study [7]. However our study placed TNFa2 by itself as the second most represented type (16.4%), preceded only by TNFa6 (18.9%).…”

“…Strong LD between TNF2 and HLA-DRB1*0301 alleles was also found in the present study, which indicates that the association of this TNF-a variation may actually not be independent. Moreover, several other polymorphisms within the TNFA gene may also confer/modify the risk for T1D, including TNFa microsatellite (Törn et al 2006) or -1031 T/C, -863 C/A, and -857 C/T SNPs (Hamaguchi et al 2000).…”

“…These SNPs have been investigated in T1DM patients from different ethnic groups [7][8][9], demonstrating considerable differences in allele frequencies among them.…”
