Polymorphisms of Tumor Necrosis Factor-Alpha and Hepatocellular Carcinoma Risk: A HuGE Systematic Review and Meta-Analysis

Abstract: This meta-analysis shows that TNFA-308G/A, TNFA-238G/A and TNFA-863C/A polymorphisms may be associated with HCC among Asians. TNFA-857C/T and TNFA-1031T/C polymorphisms were not detected to be related to the risk for HCC.

“…Our study demonstrates that microvascular invasion was a risk that increased HCC recurrence and affected the prognosis of HCC patients, which is consistent with previous findings [6,26,27]. A close relationship between cytokines and HCC has been reported in recent years [28][29][30][31]. IL6 is a crucial effector cytokine that is involved in hepatic physiology, including hepatoprotection, the acute-phase response, and mitogenesis in liver regeneration [11,32].…”

“…One meta-analysis [39] published in 2010 about Asian population assumed that TNF-␣-308G/A polymorphism conferred a higher risk for HCC. Another meta-analysis [40] published in 2011 showed that TNF-␣-308G/A polymorphism might be associated with HCC among Asians. Whereas the relationship among Chinese Han population still remains uncertain.…”

“…The TNF-␣-308G>A genetic polymorphism, as the most studied function polymorphism, has been reported to affect the function of TNF-␣ and HCC risk [13,14]. Recently, increasing studies are conducted to investigate the association of the TNF-␣-308G>A genetic polymorphism and HCC risk in Asian population [15][16][17]. However, these findings reported conflicting results, especially in China.…”

“…A meta-analysis evaluating polymorphisms in TNF-α confirmed the relationship of the -238 polymorphism with NAFLD, whereas the -308 polymorphism was not determined as a susceptible factor (76). Additionally, TNF-α G-308A, G-238A and C-863A polymorphisms are also related to HCC in Asians populations (77). It is still unclear whether, and certainly which, TNF-α polymorphism is relevant to the development of NASH.…”
