Polyphenol‐Based Nanomedicine Evokes Immune Activation for Combination Cancer Treatment

Abstract: Engineering multifunctional nanoplatforms with high therapeutic benefits has become ap romising strategy for intractable cancer treatment. Anovel polyphenol-based nanocomplex was designed to evoke highly efficacious cancer immunosurveillance while localizing therapyo nt he primary tumor and to minimize systemic side effects. This nanocomplex is prepared via metal-polyphenol coordination by encapsulating an atural polyphenol, gossypol, and an ewly synthesized polyphenol derivative,p olyethylene glycol-Chlorin e6… Show more

“…On the other hand, Zhang et al synthesized an MPN-based nanocomplex (termed PFG) via the metal-polyphenol coordination between a PEG-chlorin e6 (Ce6) polyphenol derivative, Fe 2+ , and gossypol (a natural polyphenol). [222] The PFGs could induce immunogenic cell death (ICD) in the tumor area via PDT and chemotherapy upon 660 nm laser irradiation after intravenous injection (Figure 17). Furthermore, anti-programmed cell death ligand-1 (PD-L1) antibodies were used to reinforce the PFGbased cancer immune therapy.…”

“…A MPNbased combination therapy was reported by our group, in which the Chlorin e6 was linked to a PEG-polyphenol, serving as a photosensitizer and a self-carrier at the same time. [17] Besides, a natural polyphenol, gossypol, and ferrous ions were used for the fabrication of the final PEG-Ce6-Fe 2+ -gossypol MPNs (Figure 19A). Upon 660 nm laser irradiation, these MPNs can cause significant cancer cell DNA damage and immunogenic cancer cell death (Figure 19B), which may further induce the maturation of dendritic cells and the activation of immune system (Figure 19C).…”

“…By introducing phosphotungstate and cysteine, CDT effects were ensured at both neutral and acidic pH, thus circumventing the limited CDT effect in neutral pH. [ 46 ] Other studies on Fe‐coordinated supramolecular nanoconstructs can be divided into two categories: Fe‐coordinated natural products [ 45 , 52 , 53 , 54 , 56 , 58 , 127 ] and nanoscale polymer systems. [ 47 , 55 , 57 , 128 ] All these works demonstrated the advantages and immense potential of Fe 2+/3+ coordination in constructing multifunctional supramolecular nanodrugs for cancer theranostic applications.…”
