Polyphenols Counteract Tumor Cell Chemoresistance Conferred by Multidrug Resistance Proteins

Abstract: One of the main reasons of cancer resistance to chemotherapeutic treatment is the presence of different ABC multidrug transporters in plasma membranes. The transporters extrude wide spectrum of anticancer agents out of cancer cells at the expense of energy derived from ATP-hydrolysis. Plant-origin polyphenolic compounds, mainly flavonoids and stilbenes or their synthetic derivatives, can modulate the main ABC transporters responsible for cancer drug resistance, including P-glycoprotein, MRP1 and BCRP. The rece… Show more

“…Though in vitro anticancer activity of flavonoids is known since decades, as well as their in vivo chemopreventive potential, the reversal of MDR in cancer cells by these compounds has been barely investigated (Michalak and Wesolowska, 2012). In particular, antitumor activity on TN MDA-MB-231 cells of two aglycones, namely, quercetin and kaempferol, both belonging to the flavonol group of flavonoids as rutin (a glycoside), has been previously documented (Huang et al, 2011;Rzepecka-Stojko et al, 2015).…”

“…Fourth generation inhibitors (FGI) are natural compounds or plant extracts exhibiting less cytotoxicity and better oral bioavailability. In-vitro analysis showed MDR reversal of ABC drug transporters when treated with extracts of Chinese herbal plants such as flavonoids or stilbenes [72]. Some natural compounds such as trabectedin, cytarabin, and halaven, have been approved for clinical use based on their strong MDR reversal activity by impacting on ABC drug transporters [7376].…”

“…(16)]. Several non-toxic plant natural products have also been proposed as MDR modulators, including curcumin (20), polyphenols (21), and flavonoids (22), however, these have not yet been tested in clinical trials. Given the disappointing outcome of the modulator strategy to date, it is not clear whether this line of attack will be successful.…”
