Polyphyllin I Inhibits Propionibacterium acnes-Induced IL-8 Secretion in HaCaT Cells by Downregulating the CD36/NOX1/ROS/NLRP3/IL-1β Pathway

Abstract: Acne vulgaris (AV) is a chronic skin disease involving inflammation of the pilosebaceous units. Propionibacterium acnes (P. acnes) hypercolonization is one pathogenic factor for AV. P. acnes that triggers interleukin-1β (IL-1β) by activating the pyrin domain-containing 3 protein (NLRP3) inflammasome of the NOD-like receptor family in human monocytes. Reactive oxygen species (ROS) acts as a trigger for the production of IL-8 and activates theNLRP3 inflammasome. IL-8 promotes the metastasis and multiplication of… Show more

“…Nonetheless, Resvega reduced the IL‐8 release after hydroquinone exposure also when IL‐1β was not produced. This could be linked with Resvega's antioxidative properties and is in line with findings that ROS and NADPH oxidase participate in the elevated IL‐8 release in human keratinocytes (Yang et al, 2021). Associations between ROS regulation, IL‐1β and IL‐8 were evident also in human retinal proximal tubular epithelial cells where the oxidative stress‐induced IL‐8 release was potentiated by IL‐1β treatment, whereas IL‐8 release was reduced by treatment with the antioxidant NAC (Kumar et al, 2016).…”

“…Activation of these pathways can induce the production of pro-inflammatory factors such as IL-1β and IL-8 to regulate inflammatory and infectious diseases ( Liu et al, 2020 ). Previous evidence demonstrated that IL-1β promotes the inflammatory response of P. acnes in vitro and in vivo , and IL-8 is also an essential pro-inflammatory cytokine in the pathogenesis of acne ( Yang et al, 2021 ). Therefore, we hypothesize that the elevated PC and LPC identified in our study may be involved in the pathogenesis of acne by triggering TLR2 and TLR4-mediated signaling pathways to induce the production of inflammatory factors.…”

“…Baicalin inhibits the activation of the NLRP3 inflammasome via the NF-κB and mitogen-activated protein (MAPK) pathways, thereby reducing the inflammasome induced by Cutibacterium acnes (25). Polyphyllin I inhibits ROS production, NLRP3 activation and IL-8 secretion induced by Cutibacterium acnes and also suppresses the proliferation and migration of UVB-irradiated keratinocytes (HaCaT cells) (35). Schisandrin A, B and C inhibit the secretion of IL-1β and pyroptosis in human acute leukemia-derived monocytes (THP-1) infected with Cutibacterium acnes (19).…”
