This study was aimed at exploring biological properties of PoP for their potential use as an ingredient of choice in nutraceuticals and ethnic drug formulations. Pomegranate peel extracts (PoPx), derived from hydro-alcoholic extracts were evaluated for antioxidant, antimicrobial and potential antiulcer properties. Higher levels of total phenolics (427.19 mgGAE/g of extracts) recovered from hydro-alcoholic extracts exhibited significant (P < 0.05) ferric reducing antioxidant power (FRAP) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) free radicals scavenging activity. PoPx inhibited urease activity by 97.9% and IC50 for all extracts was estimated to be in a range of 30–44.4 μM. Potent inhibitory effects of PoP extracts were observed against Bacillus subtilis, Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Salmonella typhimurium and Aspergillus niger and minimum inhibitory concentration (MIC) was recorded between 0.25–0.89 mg/mL. The study indicates that PoPx of the white rind cultivar is a potential source of active biomolecules that could further be explored as a source of bioactive compounds and therapeutic agent against various ailments.\ud Practical Applications\ud \ud The findings of this study propose PoP of white rind cultivar as valuable waste of pomegranate juice processing industry. The proposed methodology for PoPx recovery and the bioactivities reported could equally benefit the processors for low cost recovery of protective biomolecules. The study further defines admissible levels of extracts being exploited by the industry for manufacturing nutraceuticals, functional food preparations and designing ethnopharmacological drugs of choice

Stroke is the second leading cause of death worldwide, affecting about 240 people a day in South Africa and leaving survivors with residual disabilities. At the moment, there is no clinically approved neuroprotective product for stroke but the consumption of plant polyphenols has been suggested to offer some protection against stroke. In this study, we investigated the effects of long-term consumption of fermented rooibos herbal tea (FRHT) on ischemia/reperfusion (I/R)-induced brain injury in adult male Wistar rats. FRHT was administered to the animals ad libitum for 7 weeks prior to the induction of ischemic injury via a 20-minute bilateral occlusion of the common carotid arteries (BCCAO) followed by reperfusion for 24, 96 and 168 hours respectively. Neurobehavioural deficits, brain oedema, blood-brain barrier (BBB) damage, apoptosis, lipid peroxidation and total antioxidant capacity were subsequently evaluated using standard methods. Our results showed that long-term consumption of FRHT by Wistar rats significantly reduced brain oedema and neuronal apoptosis, but did not attenuate BBB damage following cerebral ischemia. Analysis of whole-brain homogenates showed significantly reduced lipid peroxidation levels, increased total antioxidant capacity and resulted in improved neurobehavioural outcomes in FRHT-treated rats when compared with untreated animals. Taken together, our results tend to suggest that continuous consumption of FRHT could confer some protection against ischemic brain injury (IBI) and is therefore highly recommended for patients with stroke-predisposing conditions.

ABSTRACT. The aim of this study was to investigate the effect of lyophilized pomegranate extract on spermatological features, pathology of testes and total antioxidant/oxidant status in rabbits. Adult male rabbits were divided into four groups containing six rabbits each. For 8 weeks, Group I received standard diet and 1 ml % 0.5 carboxymethyl cellulose (CMC), Group II received 25 mg/kg/day pomegranate extract + 1 ml % 0.5 carboxymethyl cellulose (CMC), Group III received 50 mg/kg/day pomegranate extract + 1 ml % 0.5 CMC and Group IV received 100 mg/kg/day pomegranate extract + 1 ml % 0.5 CMC. Rabbits were sacrificed by using xylazine 5 mg/kg+ketamine 35 mg/kg anaesthesia and were euthanized by 150 mg / kg intraperitoneal thiopental sodium at the end of the eighth week. Spermatozoon motility, abnormal sperm rate, sperm membrane integrity, total antioxidant/oxidant level and spermatogenic cell density were investigated. All analyses were done only once at the end of study period. Data were compared by analysis of variance (ANOVA) and the degree of significance was set at (p<0.05). Sperm motility and membrane integrity increased significantly (p<0.05) in groups II, III and IV; abnormal sperm rate decreased significantly (p<0.05) in groups III, IV; total oxidant status decreased significantly (p<0.05) in group IV in comparison to the control group. Seminiferous tubule diameter increased significantly (p<0.05) in all groups compared to the control group. Germinal cell layer thickness significantly increased (p<0.05) in group IV compared to the control group. Results of this study suggest that 50 mg/kg/day + 1 ml % 0.5 CMC and 100 mg/kg/day + 1 ml % 0.5 CMC improve sperm parameters in rabbits.
